The quality of dNTPs
Deoxynucleotide triphosphates (dNTPs) are the building blocks of DNA and are essential components in every PCR or qPCR reaction. The quality of the dNTPs has a significant impact on the PCR reaction yield, amplification accuracy and the length of the PCR product. For example, amplification of very long targets or of complex DNA templates is not possible without the highest purity dNTPs.
Impurities found in low-quality dNTPs such as chemical contaminants or incomplete synthesis products drastically affect the amplification yield and accuracy, leading to poor PCR results. To ensure reliable PCR results, it is essential to use dNTPs that have high purity of at least 99% confirmed by HPLC analysis.
The dNTPs shall be free from deaminated nucleotides, pyrophosphates, from nucleases that can damage the DNA and from proteases that can damage the enzymes. Highest quality dNTPs, such as the ones from highQu are also tested to be free from bacterial and human DNA contamination and are stable during the prolonged storage and after multiple freezing/thawing cycles.
