highQu ready to use Prestained Protein Ladders are mixtures of highly recombinant proteins with coupled chromophores providing sharp protein bands and bright colors on denaturing polyacrylamide gels. The high ladder purity allows for exceptional stability and room short term temperature storage. Ladders are ready to be directly loaded on gels without any preparation or heating. They provide sharp bands for approximate protein sizing and allow for monitoring of electrophoresis process and Western transfer efficiency.
Migration differences of protein ladder proteins in different PAA gels and buffers
It is commonly known that prestained proteins of colored protein ladders migrate differently from unstained natural proteins of the same molecular weight. Prestained protein ladders are therefore not suitable and not recommended for a precise MW estimation. Even under the denaturing SDS PAGE conditions, the chromophores of the ladder proteins influence protein migration pattern. Therefore, to get more precise MW comparison results in electrophoresis, every prestained protein ladder is previously calibrated under the same protein gel electrophoresis conditions (for example, Tris-Glycine 4-20% gradient gel and Bis-Tris 10% MES buffer) to estimate the appearing molecular weight of the protein ladder. Calibration is done by comparing migration on the same gel with the same size unstained proteins. The influence of the buffer shall be also taken into account when analyzing protein migration. For example, Tris-acetate changes protein migration significantly, compared to Tris-glycine buffer. Companies selling protein ladders do provide migration chats for prestained proteins to show approximate apparent molecular weight of the ladder under different gel conditions.
